Yes, you can invoke VaraFM or Soundmodem (packet modem) from the same Winlink instance. As long as you call them one at-a-time they’ll work fine.
Running two modem apps at the same time is not going to work because both will try to access serial port for PTT by RTS. You can work around that by virtualizing the serial port.
Can I set up two instances of winlink, one for Vara and one for soundmodem/packet, and have them running at the same time while using the same radio and antenna?
I have not seen a lot of outside usage of my station, so packet and vara conflicts would be minimal if at all.
I may be off on this, but in my mind Winlink is used in a client setup only. With that you’ll need only one of the two modem apps at-a-time depending on the mode supported by the digipeater you are connecting.
If you are setting up a digipeater, then it’s a different use case and I don’t think it involves Winlink app. Somebody with more practical experience may be able to chime in on this or you can try researching on the web/youtube how packet/vara digipeaters are setup.

I run 2 physical Winlink Gateway Stations.
1st Gateway is on HF/VHF/UHF.
On VHF I run UZ7HO Sound Modem for packet and VARA-FM using the Digirig Lite.
Digirig does not use Comports for PTT like the Mobile version. This eliminates that conflict.
2nd Gateway is only VHF/UHF and uses same setup using The Lite version.
Hoping to get up to one of our Packet Node sites to setup a VARA-FM Digipeater this year.
Like Denis said you can run both Packet and VARA-FM on the Client side but not at the same time. I hope that is as clear as MUD. LOL! 73 de Steve KB6HOH
